1. Intended Use
Varicella Zoster Virus (VZV) real time PCR kit is used for the detection of Varicella Zoster Virus in
blood, lesion swab, lesion scab or stool samples by using real time PCR systems.
2. Product Description
Varicella-zoster virus (VZV) is a DNA virus, and is a member of the human herpesvirus group, causes
varicella or chickenpox, a common infectious disease of childhood. Like other herpesviruses, VZV has
the capacity to persist in the body after the primary (first) infection as a latent infection. It persists in
sensory nerve ganglia. Primary infection with VZV results in chickenpox. Herpes zoster (shingles) is
the result of recurrent infection. VZV has a worldwide distribution, 80 % of children under 10 years
have had varicella. Varicella Zoster Virus real time PCR kit contains a specific ready-to-use system for
the detection of Varicella Zoster Virus by polymerase chain reaction (PCR) in the real-time PCR
system. The master contains reagents and enzymes for the specific amplification of VZV DNA.
Fluorescence is emitted and measured by the real time systems´ optical unit during PCR. The detection
of amplified VZV DNA fragment is performed in fluorimeter channel FAM with the fluorescent
quencher BHQ1. DNA extraction buffer is available in the kit and blood serum, lesion swab or lesion
scab samples are used for the extraction of the DNA. In addition, the kit contains a system to identify
possible PCR inhibition by measuring the HEX/VIC/JOE fluorescence of the internal control (IC). An
external positive control (1×107copies/ml) allows the determination of the gene load. For further
information, please refer to section 9.3 Quantitation.
