1. Intended Use
Leptospira real time PCR kit is used for the detection of leptospira in serum, urine or C.S.F samples by using real time PCR systems.
2. Product Description
Leptospirosis is a bacterial zoonosis that is endemic worldwide, with a higher incidence in tropical climates. It is caused by spiral-shaped bacteria of the genus Leptospira, (referred to as leptospiras) which infect a variety of wild and domestic animals. There are over 200 known pathogenic serovars, which infect different species of animals. A variety of wild and domestic animals may act as reservoirs for leptospiras, excreting the organism in their urine or fluids of parturition. Humans may be infected through direct contact with urine or fluids of parturition of infected animals, or through contact with contaminated water or soil. Leptospira real time PCR kit contains a specific ready-to-use system for the detection of the leptospira (for serotypes of L.icterohemorrhagie, L.canicola, L.ballom, L.pyrogenes, L.hebdomadis and L.pomona ) by polymerase chain reaction in the real-time PCR system. The master contains reagents and enzymes for the specific amplification of the leptospira DNA. Fluorescence is emitted and measured by the real time systems´ optical unit. The detection of amplified leptospira DNA fragment is performed in fluorimeter channel FAM with the fluorescent quencher BHQ1. DNA extraction buffer is available in the kit. In addition, the kit contains a system to identify possible PCR inhibition by measuring the HEX/VIC/JOE fluorescence of the internal control (IC). An external positive control (1×107copies/ml) contained, allows the determination of the gene load. For further information, please refer to section 9.3 Quantitation.
